Any Screen you want: Customized Optimization Screens
If you want an optimization screen from an Emerald BioSystems screen, just tell us the formulation for the condition or conditions that you want to optimize. For any lead conditions, whether from an Emerald BioSystems screen or not, we can make your desired optimization screen through several different methods using our Crystal Miner software. Standard grid screening optimization screens are made using our software's Component Variation feature. Gradients can be designed based on pH and concentrations for all components in the lead condition. Reagents not present in the lead condition can also be added to the optimization screen. The image to the left is an example of the Component Variation Optimization of Wizard condition #28 to make a new 24-condition screen.

Optimization screens can also be made by systematic quadrant optimization, shown in the table to the left. In this optimization scheme, a lead condition is used to generate four separate 24-condition screens. In each 24-condition screen, one or two components among pH, precipitants concentration, and salt concentration are varied systematically. The final optimization screen is a 96-condition screen composed of the four 24-condition screens.The lead crystallization condition must contain one precipitant, one buffer and one salt; or one precipitant, one buffer, and no salt to generate a systematic quadrant optimization screen

| Finally, optimization screens can be made from multiple lead conditions through our combinatorial PBS Cross method. In the PBS Cross method, all possible combinations of conditions with one precipitant, one buffer, and one salt are generated using all of the precipitants, buffers and salts from multiple lead conditions (at their pHs and concentrations from the lead conditions), as shown in the image below. PBS Cross optimization screens can be very useful in determining which components among several lead conditions are most important for crystallization. |
 In this example of a PBS Cross optimization screen, the lead conditions were Wizard #11, #17 and #18. Among these three different conditions are three different precipitants (blue hues), three different buffers (red hues), and two different salts (green hues). The total number of different combinations of conditions with one precipitant, one buffer, and one salt in each condition is therefore, 3x3x2=18. Additional precipitants, buffers, and salts can be added to increase the combinatorial diversity of the optimization area.
